Lead Research Specialist

GENERAL PURPOSE:The Genomics Research Center (GRC) at the University of Rochester is seeking a highly motivated and organized candidate to provide comprehensive technical support of high-throughput sequence data generation to customers of the Genomics Research Center (GRC). Our interactive team is focused on fast turn-around of high-quality results using state-of-the-art genomic methods and proper handling of large quantities of sequencing data. Our team routinely researches, creates, and refines in-house methods to achieve throughput goals while maintaining the quality and scientific integrity of deliverables for every sequencing project. This position would mainly focus on daily operations including: molecular biology, DNA and RNA isolation from human samples, DNA and RNA analysis, PCR, high-throughput sample processing and DNA sequencing.JOB DUTIES AND RESPONSIBILITIES:

Analyze gene expression in cells and the tissues utilizing qPCR and high- throughput RNA sequencing methods. Restriction digest mapping, sub- cloning, plasmid preparation, DNA and RNA isolation, poly (A)-oligo (dT) mRNA purification, polymerase chain reaction, cDNA synthesis, RT-PCR, Next Generation Sequencing (NGS) library construction, and Illumina sequencer operation.

Design, develop, support single cell omics experimentation aligned with client needs. This includes initial design and consultation, coordination of sample handoff, sample processing and quality control, single-cell capture and library construction, high-throughput sequencing, and post sequencing feedback on success.

Interpret RNA and DNA quality assurance results from GRC QA/QC service line. Data manipulation using computer assisted software such as Adobe Photoshop, Agilent Bioanalyzer 2100 EXPERT software, PowerPoint, Excel, and GraphPad PRISM. Prepare figures for manuscripts and presentations and write manuscripts. Attend local, national and international meetings to present data and learn new information. Keep abreast of current literature in fields of research. Prepare data and new experimental methods for NIH Grant applications. Prepare figures in final form for publication and write first draft of manuscripts derived from independent research projects. History of publications expected.

Direct/manage GRC cell culture operations, including training and supervision of staff on best practices and methods.

Establish standard operating protocols for nucleic acid isolation, plasmid preparation, DNA library construction, cell specific culture techniques, RNAi/CRISPR experimentation, and single-cell RNA sequencing experimental design Function as liaison between the PI’s laboratory and Environmental Health and Safety. Maintain inventory, policy awareness and enforcement of such policies. Regularly scan the literature and vendor’s catalogs to find new reagents and techniques and implement such to enhance research output.

Train and supervise professional laboratory support staff, graduate students, and postdoctoral researcher working with the core laboratories in experimental design and techniques, including the use and maintenance of instrumentation. Interpret data and direct research activities of junior technical support personnel. Provide trouble-shooting assistance.

Supervise and train others in the cell culture techniques, flow cytometry, and molecular cloning methodology.

Keeps abreast of trends in field of interest by reading professional journals; and attend/participate in weekly lab meetings, and, as appropriate, institutes and seminars.

Other duties as assigned.QUALIFICATIONS:

Master’s Degree required.

PhD preferred.

5 years of related work experience involving direct assistance to researchers or equivalent combination of education and experience required.

Unique skill set – will vary with research project need required.

Demand for skill set/research outcomes on a national/international/or corporate basis required.

Certification aligned with critical need preferred.
